多脂鳞伞多糖提取物对链脲佐菌素诱导糖尿病小鼠抗糖尿病活性Anti-diabetic effects of fungal polysaccharides extracted from Pholiota adiposa on streptozotocin-induced diabetic mice

摘要(Abstract):
多脂鳞伞Pholiota adiposa是一种珍贵的食药用真菌。本文研究了多脂鳞伞子实体中提取的真菌多糖(PAP)对高脂饮食联合链脲佐菌素诱导的2型糖尿病小鼠的抗糖尿病作用。研究表明,PAP治疗组的空腹血糖水平显著降低,体重增加,并且三酰甘油(TG)、总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)的水平降低,而超氧化物歧化酶(SOD)、高密度脂蛋白胆固醇(HDL-C)的水平、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)在PAP治疗组中显著增加。模型组的胰腺和肝脏组织表现出显著的病理学变化,PAP治疗组具有改善作用,PAP治疗组胰岛形态和功能显著恢复,此外,蛋白质印迹分析表明,PAP处理后PI3K、AKT、p-AKT、p-IRS1、IRS1、GSK3β和p-GSK3β等蛋白的表达增加,PI3K、p-AKT等信号通路与胰岛素抵抗的减轻相关。综上所述,PAP能降低小鼠空腹血糖水平、调节机体氧化应激性、保护肝脏及胰腺等器官并且能够介导PI3K、p-AKT等信号通路起到预防和治疗糖尿病的作用。
